Title: "Sparkle Boost" Question

I'm going to build Dragonfly's "Sparkle Boost" with the NTE 458 Transistor.I was wondering on the schematic it has a 100k Trimpot and the transistor.Does either one of them need to be "biased"?

Adjust the trimpot so that the voltage on the fet's drain is 4.5 volts.

Just bias it to what your ears like.  It's not a super picky circuit.
