checking transistor voltages for biasing?

hey...i was wondering, when checking a collector voltage....should signal be going thru the circuit? or ok just with the dc supply on?

it's something i was wondering about. i want to be sure i get an accurate read and all that.  thank you!

No need to plug the guitar in the input mate.
just measure without.

Even more emphatically - you do NOT want signal going through to measure the DC bias conditions. Signals mess that up. You can't trust the readings.

Do your bias readings with zero signal.
