Frequent Fog

I'd like to install a 50K trim pot at the input so it cleans up better with humbuckers. My question is, does it have an effect when the resistance is at zero, the way a pot does on a guitar (EG when you bypass the tone pot on an Esquire and the sound is much brighter)? Would it be best to use a "no load"  pot there?

If it's wired as a variable resistor (one end lug unconnected), then when the wiper is at the 'top' (zero resistance), all it really is is a piece of wire.  So no, there would be no effect when dialed out of the circuit.

And don't forget to connect the "unconnected lug" to the wiper.
Hence it is not "unconnected ".
